| Background | LILRB4A, also known as Gp49b, is a type I transmembrane inhibitory receptor belonging to the leukocyte immunoglobulin-like receptor family. It contains extracellular immunoglobulin-like domains, a single transmembrane region, and cytoplasmic immunoreceptor tyrosine-based inhibitory motifs. LILRB4A is expressed on several myeloid immune-cell populations, including monocytes, macrophages, dendritic cells, and mast cells. Following receptor engagement, its intracellular inhibitory motifs recruit phosphatases and suppress cellular activation, inflammatory signaling, cytokine production, and degranulation. LILRB4A contributes to immune homeostasis and the negative regulation of innate immune responses, making it an important target for inflammation, autoimmune disease, myeloid-cell biology, and therapeutic antibody research. |
