| Background | EPHA2, also known as EphA2 or ECK, is a type I transmembrane receptor tyrosine kinase belonging to the Eph receptor family. The extracellular region contains an ephrin-binding domain, a cysteine-rich region, and two fibronectin type III domains, while the intracellular region contains a protein tyrosine kinase domain and a sterile alpha motif. EPHA2 binds membrane-associated ephrin-A ligands, particularly ephrin-A1, and mediates contact-dependent signaling between adjacent cells. EPHA2 signaling regulates cell adhesion, migration, proliferation, differentiation, angiogenesis, and tissue development. Dysregulated expression or activation of EPHA2 has been observed in multiple cancers and is associated with tumor growth, invasion, metastasis, and treatment resistance. Therefore, EPHA2 is an important target for cancer biology, antibody development, targeted drug delivery, and immuno-oncology research. |
