| Background | Staphylococcal Sortase A is a bacterial transpeptidase that covalently links proteins to the bacterial cell wall by cleaving between threonine and glycine at an LPXTG recognition motif and catalyzes the formation of an amide bond between the carboxyl-group of threonine and the amino-group of the cell-wall peptidoglycan. This chemistry can be exploited to site-specifically link proteins/peptides with the C-terminal LPETGX motif to other proteins or molecules possessing a glycine or aminomethylene motif. |

Figure 2. To evaluate whether Sortase A 5M–mediated site-specific biotinylation preserves protein functionality, BCMA-hFc was biotinylated at the C-terminus and assessed for antigen-binding activity by ELISA. Biotinylated Human BCMA-hFc (PME101869B) shows dose-dependent binding to immobilized Human BAFF-hFc (PME100043) , as detected by HRP–streptavidin, with an EC50 of 7.03 ng/mL.
Figure 3. To assess the suitability of Sortase A 5M for antibody functionalization, anti-B7-H3 mAb (BME100010) was site-specifically biotinylated and evaluated in a sandwich ELISA format. Biotinylated anti-B7-H3 mAb (BME100010B) exhibits linear binding to ELISA-immobilized Human B7-H3 mFc-His (PME100012) within 16–80 ng/mL.

To validate the compatibility of Sortase A 5M–mediated PE conjugation with flow cytometry applications, anti-B7-H3 mAb (BME100010) was site-specifically labeled and functionally evaluated at the cellular level.
