CXCR4 recombinant proteins and antibodies
CXC chemokine receptor 4 (CXCR4) belongs to the rhodopsin-like family and contains 352 amino acids. Its structure includes an extracellular N-terminal domain, 7 transmembrane helices, three extracellular loops, three intracellular loops, and an intracellular C-terminal domain. The receptor can be present in a variety of polymerization states in the plasma membrane, including monomers, dimers, higher-order oligomers, and even nanoclusters.
CXCR4 is widely expressed and its expression can be detected in various developmental stages and tissue types. CXCR4 plays a key role in embryonic development, especially in the formation of the hematopoietic system, central nervous system, and cardiovascular system. In adults, CXCR4 is mainly expressed in hematopoietic stem cells, immune cells (such as T cells, B cells, monocytes), endothelial cells, fibroblasts, and some neurons and epithelial cells. After binding to the receptor, the main ligand of CXCR4, CXCL12 (also known as SDF-1), can activate the Gi-type G protein, thereby inhibiting the activity of adenylate cyclase (AC) and reducing the intracellular cAMP level. CXCR4 is highly expressed in a variety of tumor tissues and is closely related to tumor metastasis, infiltration and drug resistance. Therefore, CXCR4 has also become a focus of cancer research and targeted therapy.
