C3AR1/ C3AR ECD and Nanodisc Proteins

C3AR1, commonly referred to as C3aR or C3AR, is a seven-transmembrane G protein-coupled receptor activated by the complement-derived anaphylatoxin C3a. It is expressed by multiple immune and non-immune cell populations and participates in chemotaxis, degranulation, oxidative responses, cytokine production and regulation of innate immunity. C3AR1 signaling can influence inflammatory cell recruitment, vascular responses, tissue repair and interactions between the complement and adaptive immune systems. Depending on the tissue and disease context, C3a-C3AR1 signaling may promote inflammation or contribute to protective and regenerative responses. The receptor has therefore been investigated in autoimmune disease, inflammatory disorders, fibrosis, infection, neurological disease and cancer. Its ligand-dependent conformational changes, biased signaling and species-specific pharmacology make C3AR1 an important but technically demanding GPCR target.

C3AR1-directed drug discovery remains predominantly at the research and preclinical stages. Development strategies include small-molecule antagonists intended to reduce excessive complement-driven inflammation, agonists or partial agonists designed to exploit context-dependent protective signaling, peptide-based ligands and biologics targeting the receptor or its ligand pathway. Structural biology and medicinal chemistry studies are helping define orthosteric and allosteric binding pockets and distinguish productive signaling from receptor desensitization or biased pathway activation. However, commonly used research compounds may show off-target activity or different pharmacology across species, complicating translation from animal models to humans. Clinical validation of selective C3AR1 modulation remains limited, and improved assays are needed to evaluate receptor occupancy, ligand competition, signaling bias and target specificity.
