Triggering Receptor Expressed on Myeloid Cells 2 (TREM2) is a type I immunoglobulin-superfamily receptor expressed primarily on microglia, macrophages, monocytes, and osteoclasts. Its short cytoplasmic tail lacks intrinsic signaling motifs and therefore associates with DAP12/TYROBP through the transmembrane region to initiate ITAM-dependent signaling. The extracellular IgV-like domain can recognize lipids, phospholipids, apolipoproteins, and damage-associated ligands. TREM2 signaling regulates myeloid-cell survival, migration, phagocytosis, lipid metabolism, and inflammatory state. In the central nervous system, it is particularly important for microglial responses to apoptotic cells, myelin debris, and pathological protein aggregates. Disease-associated TREM2 variants have made the receptor a major target in neurodegeneration and neuroimmunology, while TREM2-positive myeloid populations are also being investigated in tumor biology.
