E-Cadherin/CDH1 Target and Research Tools for Cancer Studies

CDH1 (Cadherin-1, E-Cadherin) is one of the most extensively studied targets within the classical cadherin family and a well-established cancer-related target. Predominantly expressed on epithelial cell surfaces, CDH1 mediates calcium-dependent cell–cell adhesion via its extracellular domains, playing a central role in maintaining tissue integrity, cell polarity, and epithelial homeostasis. Loss or dysfunction of CDH1 is a hallmark of epithelial–mesenchymal transition (EMT) and is closely linked to tumor invasion, metastasis, and therapy resistance.

As a representative cadherin family target, CDH1 has attracted sustained interest in drug discovery. To date, dozens of CDH1-associated drug development programs are underway globally, spanning monoclonal antibodies, antibody drug conjugates (ADCs), and combination therapeutic strategies.
