CEACAM5 recombinant proteins and antibodies

Human carcinoembryonic antigen-related cell adhesion molecule 5 (CEACAM5) is a tumor-specific antigen first discovered in colon cancer by Gold and Freeman in 1965. CEACAM5, also known as CEA (carcinoembryonic protein) or CD66e, is the first member of the carcinoembryonic antigen-related cell adhesion molecule (CEACAMs) family, which has a total of 12 members. Compared with other CEACAMs molecules expressed in hematopoietic cells (such as CEACAM1, CEACAM8, CEACAM6, etc.), the expression of CEACAM5 has not been detected in hematopoietic cells. Related studies have shown that the expression of CEACAM5 begins in the early stages of human embryonic and fetal development (9-14 weeks) and remains expressed in certain cells throughout life. In normal adult tissues, CEACAM5 is mainly present in colon columnar epithelial cells and goblet cells, and is also present in the stomach, tongue, esophagus, cervix, sweat glands and prostate. It is worth noting that under normal circumstances, the expression of CEACAM5 in lung and gastrointestinal epithelial cells is limited to the apical surface of the epithelial cell membrane facing the cavity, so it cannot be recognized by immune cells. In cancer cells, CEACAM5 is highly expressed mainly on the surface of epithelial tumor cells, including colorectal cancer, gastric cancer, pancreatic cancer, gallbladder cancer, lung adenocarcinoma, small cell lung cancer, breast cancer, bladder cancer, ovarian cancer, etc.

CEACAM5 has always been used as a tumor marker in the clinical diagnosis and treatment of colorectal cancer to help diagnose and detect tumor development. In recent years, with the deepening of research, the potential of CEACAM5 as a tumor target for new drug development has gradually emerged. In clinical development, researchers are comprehensively developing CEACAM5 as a variety of novel therapeutic targets for biological therapies, including ADC, bispecific antibodies and chimeric antigen receptor T cell therapy.
