CLDN6 Full-Length Membrane Proteins and Antibodies

Tight junction protein 6 (CLDN6) is a four-pass transmembrane member of the claudin family and an integral component of epithelial tight junctions. Claudins regulate paracellular permeability, epithelial barrier integrity, and cell polarity through their transmembrane segments and two extracellular loops. CLDN6 is prominently expressed during embryonic development but is strongly downregulated in most normal adult tissues. Re-expression of CLDN6 has been observed in multiple solid tumors, creating a distinctive tumor-associated cell-surface expression profile that has attracted interest in monoclonal antibodies, ADCs, bispecific and multispecific molecules, and engineered cell therapies. Because CLDN6 shares substantial sequence and structural similarity with closely related claudins such as CLDN9, maintaining target selectivity while recognizing the native membrane conformation is a major consideration during antibody discovery.
