SIRPα/SIRPA/CD172a Proteins & Antibodies

SIRPα, encoded by the SIRPA gene and also known as CD172a, is an inhibitory receptor of the immunoglobulin superfamily that is predominantly expressed on myeloid cells, including macrophages, monocytes, dendritic cells, and neutrophils. The intracellular region of SIRPα contains ITIM motifs that recruit phosphatases such as SHP-1 and SHP-2 following ligand engagement. Its principal ligand, CD47, delivers an inhibitory signal through SIRPα that reduces macrophage-mediated phagocytosis. The CD47-SIRPα pathway is therefore widely studied as a major innate immune checkpoint and a key regulator of myeloid-cell activity. It has become an important area of research in tumor immunology, macrophage biology, and immune-modulatory drug development. SIRPα also exists in multiple genetic variants, and species differences can affect ligand binding and antibody recognition, making variant specificity and cross-species reactivity particularly important during biologic development.
