ADAM9 (A distegrinin and a metalloprotease 9), also known as MDC9 or meltrin-γ, is a member of the disintegrin and metalloprotease (ADAMs) family, which belongs to the zinc protein superfamily. ADAM9 exists in both mice and humans and was first discovered in breast cancer in 1996. Its protein structure is a typical ADAMs structure, consisting of an N-terminal signal peptide (1-29aa), a prodomain (30-205aa), a metal chelate hydrolase domain (206–412aa), a disintegrin domain (413-503aa), a Cys-rich domain (504-643aa), an EGF-like domain (644-698aa), a transmembrane domain (699-718aa) and a cytoplasmic domain (719-819aa). ADAM9 is widely expressed in human tissues, including lungs, colon, kidneys, vascular smooth muscle, nervous system, reproductive system, etc., and its expression increases under pathological conditions. ADAM9 is expressed in a variety of cell types, including monocytes, macrophages, neutrophils, keratinocytes and fibroblasts.
