Claudin 18.2, also called CLDN18.2, is a four-pass tight-junction membrane protein containing two extracellular loops. It is one of the major isoforms produced from the CLDN18 gene and is physiologically associated primarily with gastric mucosal epithelium. In healthy tissue, CLDN18.2 is largely confined within tight-junction structures, limiting antibody accessibility. During malignant transformation, however, disruption of epithelial polarity and junction organization can expose CLDN18.2 on the surface of tumor cells.
