DSG2 Target and Research Tools for Cancer Studies
Desmoglein-2 (DSG2), a member of the cadherin superfamily, is a core transmembrane component of desmosomes. Functioning like a “molecular anchor,” it tethers adjacent cells to provide the mechanical strength necessary for cardiac and epithelial tissues to withstand physical stress. Unlike other desmogleins confined to stratified epithelia, DSG2 is ubiquitously expressed in all desmosome-bearing tissues, with prominent levels in cardiomyocytes and simple epithelia (e.g., intestine, lung). Beyond its structural role, DSG2 also modulates signaling pathways related to apoptosis, proliferation, and angiogenesis.
Abnormalities in DSG2 are linked to various pathologies. In cardiology, mutations in the DSG2 gene are primary drivers of Arrhythmogenic Right Ventricular Cardiomyopathy (ARVC). In oncology, DSG2 plays a complex dual role: it is often upregulated in gastric cancer, melanoma, and squamous cell carcinoma, driving progression by promoting survival and vasculogenic mimicry (VM). Conversely, its downregulation is associated with increased invasiveness in certain breast and prostate cancers. Notably, DSG2 serves as a receptor for several human adenoviruses (e.g., Ad3, Ad14). Current research explores the use of DSG2-targeting “junction openers” to enhance the penetration of chemotherapeutics or monoclonal antibodies into solid tumors.
