Unlike several related receptors, HER2 has no established high-affinity soluble ligand and adopts an extracellular conformation favorable for receptor dimerization, allowing it to serve as a preferred signaling partner for other ERBB receptors. HER2-containing dimers strongly activate PI3K/AKT and RAS/MAPK signaling and regulate cell proliferation, survival, and migration. ERBB2 amplification or HER2 overexpression occurs in breast, gastric, and multiple other solid tumors, establishing HER2 as one of the most extensively studied tumor-surface targets.
