Human epidermal growth factor receptor 3 (HER3), also known as receptor tyrosine protein kinase ERBB3, is a membrane-bound protein that belongs to the ERBB/HER receptor tyrosine kinase (RTK) family. HER3 is widely expressed in human adult tissues, including cells of the gastrointestinal tract, urinary tract, respiratory tract, reproductive tract, skin, endocrine and nervous systems. Unlike other members of the ERBB receptor tyrosine kinase family, which can be activated by autophosphorylation after ligand binding, ERBB3 is kinase-deficient, with an autophosphorylation activity of only 1/1000 of that of EGFR, and has no ability to phosphorylate other proteins. Although there is no evidence that HER3 overexpression, constitutive activation or mutation itself is carcinogenic, it is associated with growth, proliferation, chemotherapy resistance, and promotion of invasion and metastasis as a dimer partner of HER2. HER3 expression or overexpression has been found in many types of tumors, such as breast cancer, ovarian cancer, lung cancer, colon cancer, pancreatic cancer, melanoma, gastric cancer, and head and neck cancer.
