KLK2 Research Tools: Antibodies and Proteins
Kallikrein-related peptidase 2, commonly known as KLK2 or hK2, is a secreted trypsin-like serine protease expressed predominantly in prostate tissue. Its transcription is regulated by androgen-receptor signaling, and elevated expression is frequently maintained in prostate cancer, including some advanced and treatment-resistant disease states. KLK2 participates in proteolytic pathways within the prostate and can activate related proenzymes involved in seminal-fluid biology. Its restricted tissue distribution and association with androgen-driven tumor cells make it an attractive biomarker and therapeutic target. Although KLK2 is secreted, a proportion can remain associated with the tumor-cell environment, allowing antibodies and other targeting agents to distinguish prostate-derived tissue from many non-prostatic organs. Patient selection, target accessibility and differences between circulating and tumor-associated antigen remain important considerations for therapeutic development.
KLK2-directed drug research includes T-cell-engaging antibodies, radioligand approaches, antibody-based payload delivery and engineered cell therapies. T-cell redirection is among the most clinically advanced modalities, with first-in-human Phase I studies reporting initial safety, pharmacodynamic and antitumor findings in advanced prostate cancer. Radiopharmaceutical strategies aim to exploit the prostate-restricted expression pattern to deliver radiation directly to KLK2-positive lesions and are being explored from preclinical development into early clinical evaluation. CAR-T and other cellular approaches remain largely preclinical or at an early translational stage. Additional concepts include antibody–drug conjugates and combination regimens designed to improve tumor penetration or address heterogeneous antigen expression. The overall field is still developing, with ongoing work focused on dose optimization, cytokine-related toxicity, antigen density and activity in heavily pretreated disease.
