LGR5 Recombinant Proteins and Flow Cyt Antibody
Leucine rich repeat containing G protein-coupled receptor 5 (LGR5) is a member of the LGR family of GPCRs, featuring a seven-transmembrane architecture together with a large leucine-rich extracellular domain. Functionally, LGR5 serves as a receptor for R-spondins, participates in Wnt/β-catenin signaling, and is closely associated with adult stem cell maintenance and cancer stem cell biology.
From a drug-development perspective, the LGR5 pipeline is still relatively selective rather than crowded, but it is clearly moving beyond basic biology. Publicly trackable programs already include at least one clinical-stage LGR5-related bispecific antibody program and one clinical CAR-T study, while preclinical and conference-stage disclosures have expanded into ADCs, bispecific antibodies, VHH/nanobody-based approaches, and CAR-T. The main disease focus remains solid tumors, especially colorectal, gastric, liver, ovarian, and brain cancers.
In fact, LGR5 is widely viewed as a target with clear biological relevance but real development complexity. On the one hand, it is strongly linked to tumor initiation, relapse, and treatment resistance. On the other hand, as a seven-pass membrane protein with a complex extracellular region, LGR5 is highly dependent on native conformation and membrane context, while its biological role in normal stem cell compartments also raises the bar for antibody discovery, binding evaluation, and translational safety assessment.
To address these challenges, DIMA BIOTECH currently offers several LGR5 products, including LGR5 ECD proteins, full-length LGR5 membrane proteins in synthetic nanodiscs, and Flow Cyt-validated reference antibody. The two hFc-tag ECD proteins are better suited as soluble antigens for immunogen preparation, ELISA coating, preliminary binding validation, and early antibody screening. The two full-length nanodisc proteins are more suitable for applications that require native-like membrane context, including conformation-dependent binding studies, membrane-proximal epitope analysis, mechanism research, and more challenging membrane-protein drug discovery workflows.
