Transmembrane proteins are vital cell membrane components involved in transport, signaling, and recognition. Their dysfunction often causes disease, making them major drug targets: over 60% of known drug targets and >90% of antibody drug targets are membrane proteins. Yet, drug development is challenging due to their hydrophobic nature, which complicates soluble expression and native conformation maintenance. Full-length multi-span proteins are usually expressed at low levels, further hindering antigen preparation. Successful antibody development depends on obtaining membrane proteins in their native, functional state. Current antigen strategies and challenges include:

• • Extracellular domain (ECD) expression – simple, high yield, but may lose conformational epitopes.
  • Whole-cell immunization – preserves native conformation and modifications but risks nonspecific antibodies due to complex cell components.
  • Full-length proteins – ideal antigens but difficult to express, isolate, and purify.
