C-C chemokine receptor type 8 (CCR8), also known as CDw198, CKRL1, CMKBR8, or CMKBRL2, is a class A chemokine GPCR with a canonical seven-transmembrane architecture. CCR8 responds primarily to CCL1 and regulates Gi-dependent calcium signaling, chemotaxis, and immune-cell migration. Physiologically, it contributes to Th2-cell recruitment and skin and mucosal immune homeostasis. In cancer, CCR8 is particularly notable for its enrichment on tumor-infiltrating regulatory T cells while remaining comparatively low on peripheral Tregs and many normal tissues. This expression pattern has made CCR8 a highly attractive target for selectively modulating or depleting immunosuppressive intratumoral Tregs.
