Gastric inhibitory polypeptide receptor (GIPR) belongs to the B1 class receptor family of GPCR and can bind to glucagon-like peptide (GLP). GIPR protein contains a large extracellular N-terminal domain, seven transmembrane domains and a short intracellular C-terminal domain. GIPR is widely distributed in the body, including the pancreas, stomach, small intestine, adipose tissue, heart, and brain tissue. It is mainly present in pancreatic β cells. GIPR activated by GIP binds to the heterotrimer Gs (αβγ), induces adenylate cyclase activation, increases cAMP levels in the cytoplasm, and cAMP activates PKA, phosphorylates regulatory gene transcription proteins, and causes them to translocate to the nucleus.
