G Protein-Coupled Receptor 75 (GPR75), a member of the GPCR family, was first identified by Tarttelin et al. in 1999. It plays a significant role in various diseases, including obesity, cancer, and metabolic syndrome, making it one of the most promising targets for weight loss therapy.
GPR75’s emergence as a novel target for obesity treatment is primarily based on a groundbreaking study published by Regeneron in the journal Science in July 2021. This study analyzed the relationship between rare genetic mutations and body mass index (BMI) by conducting whole-exome sequencing on 645,000 individuals from the UK, USA, and Mexico. The researchers identified 16 nonsynonymous genetic mutations associated with BMI. Notably, they found that truncating GPR75 mutations occurred at a rate of 0.004%, with heterozygous mutations leading to a lower BMI by 1.8 kg/m², a weight reduction of 5.4 kg, and a 54% lower risk of obesity. Additionally, in vivo validation studies in mice demonstrated that GPR75 knockout mice had lower body weight, blood glucose levels, and insulin levels.
