Nectin-4 recombinant proteins and antibodies

Nectin-4, also known as poliovirus receptor-related protein 4 (PVRL4), is a transmembrane protein that belongs to the Nectins family of immunoglobulin-like molecules. This family also has three members: Nectin-1, Nectin-2 and Nectin-3, all of which are single-pass transmembrane proteins and belong to Ca 2+-dependent immunoglobulin-like molecules. Nectin-4 is a transmembrane cell adhesion molecule with a molecular mass of about 66 kDa encoded by the NECTIN4 gene. In normal tissues, Nectin-1, -2 and -3 are widely expressed in the body, including epithelial cells, neurons and fibroblasts, and nectin2 and 3 are also expressed in cells that do not express cadherin, such as blood cells and sperm cells. Unlike the widespread expression of Nectin-1, -2 and -3, Nectin-4 is widely enriched in embryonic and placental tissues, including skin, tonsils and tubular structures (trachea, esophagus and nasopharynx), and its level is significantly reduced in adulthood. In recent years, nectin-4 has been found to be overexpressed in a variety of malignant tumors, including breast cancer, ovarian cancer, colorectal cancer (CRC), prostate cancer, and lung cancer. In addition to the membranous form, serum nectin-4 levels are significantly increased in patients with non-small cell lung cancer (NSCLC) or serous ovarian cancer. Currently, there is only one ADC drug targeting nectin-4 on the market in the world, namely enfortumab vedotin, and there are 8 nectin-4 ADC drugs that have entered the clinical stage.
