Delta-Like Ligand 3 (DLL3) is a single-pass transmembrane protein attached to the cell surface. Together with DLL1 and DLL4, it forms the Delta-like ligand (DLLs) family. DLL3 is an inhibitory ligand of the Notch signaling pathway [3] and plays a key role in Notch signaling. The Notch pathway is a highly conserved intercellular signaling pathway that involves multiple processes in growth and development, such as differentiation of multipotent progenitor cells, apoptosis, cell proliferation, and the formation of cell boundaries. DLL3 RNA is mainly expressed in the brain, endocrine tissues, and blood; DLL3 alone is almost not expressed in normal tissues. Studies have found that DLL3 is highly expressed in approximately 80% of small cell lung cancer (SCLC) and other neuroendocrine tumors. The differential expression of DLL3 in normal cells and tumor cells makes it an attractive tumor-selective therapeutic target. Currently, there are 16 therapies targeting DLL3, of which 15 are in the clinical stage and 1 is in the preclinical stage. The types of therapies are diverse, including antibody-drug conjuncts (ADC), CAR-T, proteins, and antibody drugs.
