SCN2A/NaV1.2 Full-Length Membrane Protein Nanodisc and Antibody
Sodium voltage-gated channel alpha subunit 2 (SCN2A) is a gene encoding ion channel protein named NaV1.2. As a major voltage-gated sodium channel alpha subunit in the central nervous system, NaV1.2 plays a central role in action potential initiation, propagation, and neuronal excitability, making SCN2A a highly relevant ion channel target in epilepsy and neurodevelopmental disorder research.
From a therapeutic perspective, SCN2A-targeted drug development is still in an early to mid-early precision neurology stage. Currently, there are 26 SCN2A-related drug programs worldwide, spanning small molecules, antisense oligonucleotides (ASOs)/individualized oligonucleotide therapies, and gene therapy, AAV, or gene-editing approaches. Of these, 4 are in active clinical development, 13 are in preclinical development, and 9 have been discontinued or show no clear progress. The main disease areas include early-onset epilepsy, SCN2A-related developmental and epileptic encephalopathy (DEE), infantile seizure disorders, autism spectrum disorder, and intellectual disability. As a typical multipass ion channel, NaV1.2 is structurally complex, difficult to express and purify, and highly dependent on membrane context for maintaining its native conformation. This makes it one of the more challenging neuronal ion channel targets in both basic research and drug discovery. For targets of this type, high-quality antigens that preserve native-like structural features are essential for antibody discovery, binding studies, structural biology, functional characterization, and therapeutic screening. To address this need, DIMA BIOTECH has launched SCN2A nanodisc, which is full-length SCN2A membrane protein reconstituted in synthetic nanodisc. By preserving membrane-associated features and native-like conformation, this format is well suited for ion channel mechanism studies, epitope and binding-site analysis, antibody screening, compound screening, and Cryo-EM-related applications. In parallel, we are also advancing Flow Cyt antibody screening for NaV1.2 through our proprietary single B cell antibody discovery platform, supporting future antibody discovery efforts for this target.
